肺腺癌与JNK研究的知识发现

        JNK信号转导通路是MAPK通路的一重要分支,它在细胞周期、生产、凋亡和细胞应激等多种生理和病理过程中起重要作用。 JNK信号转导通路是1991年发现的一类MAPK通路。

  JNK蛋白激酶由3个基因编码,jnk 1和jnk 2基因在全身广泛表达,而jnk 3呈限制性表达,仅见于脑、心脏和睾丸 J。在未受刺激的细胞中,JNK主要存在于胞质内,但核内也有一定分布。在受到刺激后, K迅速而显著地聚积于核内,并导致相应基因表达改变。JNK基因通过选择性剪接而产生10种JNK形式,JNK基因编码的蛋白具有或无一COOH末端,结果产生46 kDa和54 kDa两种蛋白。第二种剪接是选择性编码JNK功能区两个外显子中的一个,但只限于jnk 1和jnk 2基因。不同组织发出不同指令,通过选择性剪接,JNK改变停泊位点与底物的结合能力,从而决定作用底物的特异性 。
