Xuefeng Pan Model of Trinucleotide Repeats Instability (2004-2014)
Transcription → Non-B / R-loop Misfold → Gratuitous Repair / R-loop Induced DNA Replication → Repeat Instability
Author: Xuefeng Pan
This document establishes factual priority for the Xuefeng Pan Model / Pan Mechanism based on searchable, downloadable, DOI/ISBN-indexed publications.
The Model (in author's own definition)
Active transcription through trinucleotide repeat (TNR) DNA leads to formation of non-B secondary structure / R-loop (DNA misfold). Although the DNA is chemically undamaged, the misfolded structure is recognized by cellular DNA repair / replication machinery as a lesion, leading to gratuitous repair on undamaged DNA misfold / R-loop induced local DNA replication and DNA rearrangement, resulting in TNR expansion, contraction, or gene silencing.
Two specific mechanistic phrases introduced by the author:
"The Gratuitous Repair on Undamaged DNA Misfold" (title of 2011 book chapter)
"R-loop induced DNA replication in mediating trinucleotide repeat expansions" (2014 review)
