Rosa26 is a genomic mouse locus commonly used to knock-in cDNA constructs for ubiquitous or conditional gene expression in transgenic mice. 

The human homolog of the ROSA26 locus has been identified. ROSA stands for Reverse Orientation Splice Acceptor, named after the lentivirus genetrap vector.

与传统制作基因敲入的小鼠(受精卵原核注射：质粒在染色体上的整合位点不同、拷贝数不同，不同品系的异质性，小鼠传代后，转基因的拷贝数稀释以及基因沉默等原因，F0的表型可能会有不同程度的丢失)相比，Rosa26位点基因敲入小鼠模型，可以1、确定外源基因的重组位置（Rosa26基因有2个外显子，位于2个外显子之间）， 2、只有一个拷贝的插入（heterozygous），3、结合Cre-LoxP，可以使外源基因在特定组织和特定时间表达。

其实Rosa26的功能和AAVS1的功能挺像的，都是safe harbor gene, 进行genetic manipunation没有问题。
