1) 耐药性细菌感染被列为国家十大重大疾病,也是世界范围内的临床治疗难题。大环内酯类抗生素(红霉素、克拉霉素、阿奇霉素等)通过与细菌核糖体的23S RNA结合,阻断细菌蛋白质的合成释放通道达到抑菌作用。但细菌通过甲基化转移酶修饰抗菌靶点、外排泵蛋白泵出药物等方式获得耐药性。我们课题组通过GOLD和Amber等软件建模发现了药物与细菌rRNA 的新作用靶点和新作用模式(与Purdue大学合作),并利用有机化学的新方法新技术修饰大环内酯分子,以增强与rRNA的结合力;通过体外、体内抗菌实验证明候选化合物的抗敏感菌和抗耐药菌活性(与北京大学医学部、协和医学院等合作)。我们目前的研究兴趣包括我们自己课题组命名的烃基内酯(alkylide)系列、以及热门的酮内酯(ketolide)和酰内酯(acylide)系列,当前设计的重点是多靶点抗菌分子。
2)治疗老年痴呆病目前的靶点多有不足,我们课题组全合成天然产物来源的类似物,通过全新作用机制提供一条治疗老年痴呆病的新途径。
