Protective Effects of Hydrogen Gas on Renal Ischemic/Reperfusion Injury in Rats Wei Huang, M.D., Kui Zeng, M.B. Anesthesiology, West China Second University Hospital, Sichuan University, Chengdu, Sichuan, China Background Renal Ischemic/reperfusion injury was the main cause of renal dysfunction. Hydrogen gas can markedly reduce ischemic/reperfusion injury in liver and brain by inhibiting the products of oxygen free radical. This study was to determine whether inhalation of hydrogen gas could attenuate renal ischemic/reperfusion injury in rats. Methods Eighteen male Sprague-Dawley rats were randomized into sham group, control group and hydrogen gas group respectively. The right kidneys were removed in all subjects. There was no occlusion of left renal vessels in sham group. The left renal vessels were occluded for 40 min in control group. The left renal vessels were occluded for 40 min in hydrogen gas group. 2.5% hydrogen gas was inhalated 10 min before reperfusion and 120 min during reperfusion in hydrogen gas group. Plasma creatinine (Cr), blood urea nitrogen (BUN) and left kidney histological examination were measured 24 h after operation in all subjects. Results Compared to the sham group, occlusion of the left renal artery for 40 min significantly increase blood urea nitrogen (10.30±1.57 mmol/L vs.17.0±6.1 mmol/L and 38.53±7.03 mmol/L) and plasma Plasma creatinine (26.32±1.88μmol/L vs. 264.95±16.55 μmol/L and 54.55±19.76 μmol/L) in hydrogen gas group and control group. Compared to the control group, inhalation of hydrogen gas (2.5%) markedly reduced blood urea nitrogen (17.03±6.13 mmol/L vs. 38.53±7.03 mmol/L), and plasma creatinine (264.95±16.55 μmol/L vs. 54.55±19.76 μmol/L). There was no significant difference between sham group and hydrogen gas group in histological examination. The left kidney histological examination in sham group and hydrogen gas group both better than control group. Conclusion Inhalation of 2.5% hydrogen gas could attenuate renal ischemic/reperfusion injury in rats. [Key words] Kidney; Ischemic/reperfusion injury; Rats; Hydrogen gas.
