Yu and his colleagues presented a randomized clinical trial of omicron XBB.1.5-containing bivalent COVID-19 mRNA vaccine in head-to-head comparison with other two mRNA vaccines encoding earlier variants, clarifying that newer variant-matched vaccine elicits an enhanced immune response against newly emerged variants. •A newly isolated human neutralizing monoclonal antibody 39 (mAb-39) can neutralize SARS-CoV-2 and its variants.
•mAb-39 improves the activity of anti-receptor-binding domain (anti-RBD) antibody against Omicron variants.
•mAb-39 binds to a conserved epitope within the upstream region of the heptad repeat 2 (HR2) motif of Omicron spike protein.
•Point mutation within the upstream region of HR2 reduces the capacity of spike protein-induced membrane fusion.
