生物活性:DY131 (GSK 9089) 是一种有效的选择性 ERRγ 和 ERRβ 激动剂。 DY131 对 ERRα、ERα 和 ERβ[1][2] 无活性。 DY131 还抑制 Smo 信号[3]。
体外:DY131(0.1-30 μM;5 天)处理以剂量依赖性方式抑制 LNCaP-ERRγ 和 LNCaP 细胞中的细胞增殖并减少 BrdUrd 阳性细胞,对 LNCaP-ERRγ 克隆具有更高的抑制[1]< /sup>.DY131 抑制 Shh 诱导的 Smo::EGFP 积累,IC50 为 0.8 μM。 DY131 抑制 SAG (100 nM) 诱导的初级纤毛中 Smo::EGFP 的积累和 Gli 转录活性,IC50 为~2 μM[3]。DY131 显着降低由 Shh 诱导的磷酸化组蛋白 H3 (pH3) 标记的 CGNP 增殖[3]。选择性 ERRγ 激动剂 DY131 抑制 ERα 阳性子宫内膜癌的生长ERα 阴性癌细胞[4]。
体内:DY131(5 μg/kg;皮下注射;每隔一天;持续 12 天)治疗增加 P450 侧链裂解 (P450scc)、StAR 和 HMGCoA 还原酶 (HMGCR),同时降低激素敏感性脂肪酶 (HSL) 表达[5 ]。
