Today, I found one interesting research letter from Nature about stoma development.
As we know, some genes related to stoma development have been identified, most based on mutants screen, like TMM, YODA, SDD1, ERL1 and SPEECHLESS. This gene STOMAGEN, however was found based on reverse genetics. The authors first found the correlated gene with TMM, SDD1 and ER using the microarray data. I do think the authors have done many over-expression and knock-out lines of these correlated genes and found/just found this gene mutation with a obviously phenotype.
Since gene knock out may not simply result in a visible phenotype, this is important. What is more, this gene is gain of function. If u over-express stomagen or exogenous supply with stomagen peptide artificial synthesized, u will see more stoma. The authors have done many transgenic lines and got the static data.
An interesting fact is the expression of this gene is restricted in inner tissues and then sort out to the outer mature tissue or organs. It seems that this gene is important in early stage of stoma development!
Then as a traditional result, the authors checked TMM epistatic to this gene.
Significance: This study provides evidence of a mesophyll-derived positive regulator of stomatal density. Our findings provide a conceptual advancement in understanding stomatal development: inner photosynthetic tissues optimize their function by regulating stomatal density in the epidermis for efficient uptake of CO2-- from abstract of this paper!~
